I had yellow spots this year and did a lot of reading through the pumpkin archives. The two popular responses seem to be 1) Genetics or 2) Over-fertilizing (too much nitrogen). Well in my case I was feeding them way to much, so I stopped for a few weeks and I did not see any more yellow spots. My plants were about a month old, by the way.

I also had a few yellow spots on one of my culled transplants. I thought that I had accidently let a few diazanon granules rest on the leaves? Don't know

Of course, my yellow spots may have gone away anyway, without me changing anything. It's hard to say for sure, but the plant is doing great now. Mine were also accompanied by slight yellow streaks in the stalks of the affected leaves, which have not appeared on any new growth.
