I have read that you do not count the day the pumpkin was pollenated as day 1 - but instead start counting with day 1 the day after the pumpkin was pollenated. Is this the proper way?

Yes. The day of pollination is Day Zero
The following morning is Day One or 1DAP (Days After Pollination)

once you hand pollinate, do you start seeing good size gains right away??

Jeremy ususally in the first 1-3 days the fruit only swells a little but by 1 week you expect to see something a little bigger than a grapefruit. 10 days you want 25-30 inches in circumference.

is that the only sign of it being pollinated correctly or does the flower shrivle up also?

flower will always shrivel, so just look for growth, as long as it is shiny it is still good
