I noticed that on my 394.5 Vincent plant, there are three cotyledons. There are the normal 2 large ones, then one small one coming up between them on a stem similar to the first true leaf. Opposite of the 3rd cotyledon is the first true leaf starting to form. Has this mutation happened to anyone else? And is there any advantage or disadvante to it?

This may get interesting. Stand by and watch closely. I would go with it unless it becomes an unmanageable mutation inclined grower. Cottys only support the establishment of the total plant getting ready, to do it's thing.

Then again you could have a plant that might become an issue, for Rippy's Believe It or Not.

It also appears to have 2 first true leaves, does this indicate a double vine?

It seems a bit early to be guessing. Give it another two weeks or a tad more. Plant one beside it. Cut...do not pull up the one you wish to keep. Two a foot apart will not hurt each other for three or four weeks at this point.
