If I added a couple inches of leaves and topped that off with manure in the fall. what is the best way to achieve an accurate soil test in the spring? Do I use a rototiller and mix it all in and get a test, or mix it in early and wait awhile to do the test?

Jeremy, just follow the labs advice and go from there.

I take random samples from spots in the patch from a minimum of 6 inches down (or deeper).

You can go ahead and till it in .the lab will screen off the large particles as part of their soil prep

When taking soil sample -- use all plastic tools, using
metal, will show up in test.
I use hard plastic pipe, drive it down 6-8" in several different spots, then mix all, lay out on plastic, to let dry. When dry send or take to lab
